Over the last few years there has been increasing recognition that determining free drug concentrations and not total drug concentrations at or close to the site of action is a key element in predicting compound efficacy. Understanding the factors driving compound free fraction and consequently the free concentration has therefore become an essential part of a lead optimisation strategy. This seminar will highlight just some of the uses that can be made of brain tissue binding and derived free concentrations in designing compounds for CNS targets and conversely reducing the risk of unwanted CNS side effects. Translation of tissue binding to man will also be discussed.
Mass spectrometry continues to extend its role in drug discovery with the recent development of MALDI/TOF/TOF for simultaneous imaging of distribution in tissues of not only a small molecule but also its metabolites and protein biomarkers. This powerful technique will be reviewed with some examples from NiKem’s labs.
